Testing chromatin-bound metabolic enzymes as vulnerabilities in acute myeloid leukemia
Rationale and objectives:
Metabolic dependencies in the nuclear compartment can provide targetable vulnerabilities of cancer cells that can be exploited for the development of new therapeutic strategies.
Focusing on AML, we plan:
• To identify chromatin-associated metabolic enzymes using mass spectrometry on purified chromatin fractions.
• To test identified enzymes as AML-specific dependencies using pharmacologic and genetic inhibitions.
• To use targeted nuclear degradation to dissect the molecular function of selected nuclear enzymes
Envisioned Secondments
CRG-CERCA (Sdelci), months 12-15 (3 months), to receive training in the purification and mass spectrometric analysis of chromatin fractions. EISBACH, months 32-34 (2 months) to carry out a HTS compound screen and design novel drugs.

The estimated annual gross salary for the fellow will be €33.000 without family allowance; €39.000 with family allowance (approx.).
