Uncovering the role of nuclear acetyl-CoA metabolism in differentiation
Rationale and objectives:
We need to understand the function of metabolic enzymes within the nucleus. Focusing on metabolic enzymes linked to acetyl-CoA production (e.g. Acly, Acss2, Pdha) and thus fueling histone acetylation
Therefore, we plan:
• To develop a nuclear degron system for rapid and targeted depletion of metabolic enzymes only within the nucleus.
• To identify histone marks and genes regulated by nuclear metabolic enzymes using quantitative and epigenomic methods.
• To identify the biological function of nuclear metabolism by using nuclear degradation in differentiation assays.
Envisioned Secondments
UNIMI (Bonaldi), months 18-24 (6 months) to receive training in stable isotope labelling coupled to histone PTM by MS to measure which fraction of histone acetylation is dependent on these enzymes and which regions are targeted.
Recruitment
The estimated annual gross salary for the fellow will be €60.000 without family allowance; €67.000 with family allowance (approx.).
