Visualizing acyl-CoA and associated metabolites with subcellular resolution

Rationale and objectives:

We need to understand how metabolites, in particular acyl-CoAs, spatially organize in cells and their nucleus to enable rationalizing how metabolites impact on chromatin and cancer-promoting gene regulation.

Therefore, we plan:
• To develop mass spectrometry imaging (MSI) methods that allow to track acyl-CoAs with subcellular resolution.
• To combine MSI with fluorescence microscopy tools in multimodal imaging approaches to confirm/assign metabolite location.
• To use stable isotope labeling of essential nutrients in cell culture (SILEC) to visualize metabolic turnover within cell nuclei

Envisioned Secondments

TFS (Strupat) months 13-14 (2 months) to be trained in high performance LC-MS/MS of metabolites on state-of-the-art instrumentation; BI (Trefely), months 20-24 (4 months) to train in SILEC-SF and obtain reference values for MSI.

The estimated annual gross salary for the fellow will be € without family allowance; € with family allowance (approx.).
